在我国腹泻患儿中发现诺瓦克样病霉感染

诺瓦克病毒是引起无菌性急性胃肠炎爆发的重要病原。１９９０年１０月至１９９１年１月从河南省急性腹泻门诊患儿便样中发现了诺瓦克样病毒。经电镜观察,病毒直径约为２８ｎｍ,病毒壳似由有结构的亚单位组成,进一步用诺瓦克病毒特异的寡核苷酸引物做逆转录一聚合酶链式反应（ＲＴ－ＰＣＲ）,检定为阳性。由ＰＣＲ产物测得的核苷酸序列与诺瓦克病毒原型株相应序列比较,同源性为７２％。以上结果证实,在我国腹泻病人中有诺瓦克样病毒感染。这对我国病毒性胃肠炎流行的防治研究具有重要意义。     

Phosphorylation of glucose by a guanosine-5′-triphosphate (GTP)-dependent glucokinase in Fibrobacter succinogenes subsp. succinogenes S85

Cell extracts of Fibrobacter succinogenes subsp. succinogenes S85 phosphorylated glucose with a GTP-dependent glucokinase. The enzyme showed little activity with ATP (12% of that with GTP). Of other phosphate donors tested, only dGTP and ITP gave high glucokinase activities. Dialyzed extracts required Mg⁺² and K⁺ for maximal activity. In potassium phosphate buffer, glucokinase showed maximum activity at pH 7.5 with glucose-6-phosphate dehydrogenase as the coupling enzyme. In this assay, glucokinase was active with glucose (100%), 2-deoxy-d-glucose (40%), and mannose (20%). Partially purified glucokinase had a molecular weight of 82,000 and a pl of 4.82. Double-reciprocal plots of substrate concentration versus velocity were linear and the enzyme had apparent K_m values of 55 M for glucose and 72 M for GTP. Dialyzed cell extracts of Fibrobacter intestinalis C1A also contained a GTP-dependent glucokinase that showed little activity with ATP. Potassium also stimulated the activity of this enzyme. Differences in diet and behaviour of sympatric saithe and pollack in a Scottish sea loch

Two closely related and morphologically similar gadoid predators, saithe, Pollachius virens and pollack P. pollachius , coexist in close proximity on a submerged reef in Loch Ewe, Scotland. The degree of overlap between the niches of these two gadoids in the wild was investigated by means of acoustic tracking, underwater television and an examination of stomach contents. Simultaneous tracking of individuals of both species revealed that pollack generally swim more slowly than saithe, restricting much of their movements to the submerged reef. Saithe ranged more widely around the reef as part of a school during the day, moving onto the reef at night. Video recordings showed that saithe swam actively and foraged in small groups and took prey items from the kelp, whereas pollack tended to remain solitary, maintained station at particular locations for minutes at a time and apparently used the kelp forest exclusively for cover. Although the dietary overlap of the two predators was considerable, their intake of different prey groups varied. In particular, within the crustaceans, saithe took amphipods, while pollack took mysids. In addition, saithe consume a wider range of prey than pollack. The relationship between the movement patterns and the use of food resources by these two predators is discussed, with particular emphasis on differences in feeding strategies.     

Functional mitochondria in snake Bothrops alternatus erythrocytes and modulation of HbO2 affinity by mitochondrial ATP

Digitonin was applied to permeabilize the plasma membrane of Bothrops alternatus erythrocytes to study respiration, oxidative phosphorylation and Ca²⁺ transport by mitochondria in situ. These mitochondria oxidized added NAD-linked substrates, succinate and N,N,N, N-tetramethyl-p-phenylenediamine. Respiration was sensitive to rotenone and cyanide but not to antimycin A. This indicates that Bothrops mitochondria possess the respiratory complexes NADH-ubiquinone, succinate-ubiquinone, and ferrocytochrome c-oxygen oxidoreductases, although the lack of sensitivity to antimycin A raises doubt about the composition of the ubiquinol cytochrome c-reductase complex. An ability to build up and sustain a membrane potential was documented by their capacity to accumulate tetraphenylphosphonium and Ca²⁺ through an uncoupler-sensitive mechanism. Addition of ADP caused a transient decrease in the membrane potential, indicating that this is the predominant driving force for ATP synthesis as in most types of mitochondria. Structure and function of a mating-type gene from the homothallic species Neurospora africana

The homothallic Neurospora species, N. africana, contains sequences that hybridize to the A but not to a mating-type sequences of the heterothallic species N. crassa. In this study, the N. africana mating-type gene, mt A-1, was cloned, sequenced and its function analyzed in N. crassa. Although N. africana does not mate in a heterothallic manner, its mt A-1 gene functions as a mating activator in N. crassa. In addition, the N. africana mt A-1 gene confers mating type-associated vegetative incompatibility in N. crassa. DNA sequence analysis shows that the N. africana mt A-1 open reading frame (ORF) is 93% identical to that of N. crassa mt A-1. The mt A-1 ORF of N. africana contains no stop codons and was detected as a cDNA which is processed in a similar manner to mt A-1 of N. crassa. By DNA blot and orthogonal field agarose gel electrophoretic analysis, it is shown that the composition and location of the mating-type locus and the organization of the mating-type chromosome of N. africana are similar to that of N. crassa.     

The Effects of Aluminum on the Influx of Calcium,Potassium, Ammonium,Nitrate, and Phosphate in an Aluminum-Sensitive Cultivar of Barley (Hordeum vulgare L.)

The mechanism by which aluminum interferes with ion influx is not known. In this study, the effects of aluminum on the influx of the cations calcium, potassium, and ammonium and the anions nitrate and phosphate were measured in an aluminum-sensitive cultivar of barley (Hordeum vulgare L.). Aluminum (100 [mu]M) was found to inhibit the influx of the cations calcium (69%), ammonium (40%), and potassium (13%) and enhancing the influx of the anions nitrate (44%) and phosphate (17%). Aluminum interfered with the binding of the cations in the cell wall by the same order of magnitude as their respective influxes, whereas phosphate binding was strongly enhanced. The results are consistent with a mechanism whereby aluminum binds to the plasma membrane phospholipids, forming a positively charged layer that influences ion movement to the binding sites of the transport proteins. A positive charge layer would retard the movement of cations and increase the movement of anions to the plasma membrane in proportion to the charges carried by these ions.
